Overcoming Challenging ADME Studies: When and Where to Apply Accelerator Mass Spectrometry

Human radiolabeled studies provide a broad understanding of the distribution and metabolism of novel compounds.  Traditional liquid scintillation methods often provide the required detection for these studies.  In certain studies, however, the amount of radioisotope in the samples falls below the reliable detection limit of liquid scintillation counting.  This can occur if (1) the administered dose is in the microgram range, (2) the bioavailability is low, (3) the compound has low specific activity, and (4) there is a need to lower the radiolabel exposure to study subjects. In such cases, Accelerator Mass Spectrometry (AMS) is employed to overcome current detection limits, expanding the detection range to well below that of any other bioanalytical platform
